广枣叶总黄酮对心肌缺血再灌注损伤模型大鼠的保护作用及其作用机制

摘要


目的研究广枣叶总黄酮对大鼠心肌缺血再灌注损伤的保护作用与其可能存在的作用机制,明确广枣叶总黄酮的药用价值。

方法将50只SD雄性大鼠随机分为手术组和伪手术组,伪手术组给予0.9%生理盐水(20ml/kg);手术组分为对照生理盐水组(20ml/kg),阳性药物(维拉帕米)对照组(0.02g/kg),广枣叶总黄酮大剂量组(0.4g/kg),广枣叶总黄酮小剂量组(0.1g/kg)。每组10只,每日分别进行灌胃。一周后通过采用夹闭冠状动脉左前降支进行大鼠MIRI模型构造,并使用BL-420生物机能实验系统观测大鼠心律失常发生情况, TTC染色称重缺血心肌梗死重量,HE染色观察心肌组织形态学变化,酶联免疫分析法测定血清中肌酸激酶(CK)活性及血清中白细胞介素6(IL-6)、肿瘤坏死因子a(TNF-a)、核转录因子-Kβ(NF-Kβ)及一氧化氮(NO)的含量。

结果与假手术组比较,模型组大鼠心律失常的现象严重,心肌梗死灶明显,心肌组织出现明显病理损伤,血清中CK活性及TNF-a、NF-Kβ、IL-6含量明显升高(P<0.05或P<0.01),NO含量明显降低(P<0.05)。与模型组比较,广枣叶总黄酮各剂量组及维拉帕米组大鼠心律失常现象减轻,心肌梗死灶减小,心肌组织病理损伤改善,血清中CK活性及TNF-a、IL-6、NF-Kβ含量明显降低,NO含量明显升高(P<0.01)。

结论广枣叶总黄酮对大鼠心肌缺血再灌注有保护作用,其机制可能与减少心肌细胞心肌酶的漏出、减少氧自由基、抗炎作用、促进心肌和内皮细胞恢复有关。

ABSTRACT


Objective:To study the protective effect of (TFLC) on myocardial ischemia-reperfusion injury in rats and its possible mechanism. To determine the medicinal value of total flavonoids in the Chinese jujube leaf.

Methods:Fifty male Sprague-Dawley rats were randomly divided into surgery group and sham operation group. Normal saline (20ml/kg) was given in the sham operation group and the control group was normal saline group (20ml/kg), positive drug (Verapamil) control group (0.02g/kg), TFLC high-dose group (0.4g/kg), TFLC small dose group (0.1g/ Kg) .10 mice in each group were intragastrically administered. One week later, the rat MIRI model was constructed by clipping the left anterior descending coronary artery, and the occurrence of arrhythmia in rats was observed using the BL-420 biological function test system; myocardial infarct size was weighed by TTC staining; myocardial tissue was observed by HE staining. The serum creatine kinase (CK) ,interleukin-6 (IL-6), tumor necrosis factor a (TNF-a), nuclear transcription factor-Kβ (NF-Kβ) activity and nitric oxide (NO) content were determined by enzyme-linked immunosorbent assay.

Results Compared with the sham-operated group, the arrhythmia was severe in the model group, the myocardial infarct size was obvious, the myocardial tissue showed obvious pathological damage, and the serum CK activity and TNF-a, NF-Kβ levels were all increased (P<0.05),IL-6 content increased significantly (P<0.01) and NO content decreased significantly (P<0.05). Compared with the model group, TFLC and verapamil group reduced the occurrence of arrhythmia, reduced myocardial infarct size, improved myocardial tissue pathology damage, serum CK activity and TNF-a, IL-6 The contents of NF-Kβ and NF-Kβ were significantly decreased, and the content of NO was significantly increased (P<0.01).

Conclusion   There is a protective effect of TFLC on myocardial ischemia and reperfusion in rats. The mechanism is related to the reduction of myocardial enzyme leakage, reduction of oxygen free radicals, anti-inflammatory effects, and promotion of myocardial and endothelial cell recovery.
