SIRT1在抑郁症中的作用及其可能机制

摘要

抑郁症是一种常见的精神疾病,其病因、机制未完全阐明,导致临床治疗效果不佳。沉默信息调节因子2相关酶1(Sirtuin1,SIRT1)是一种NAD+依赖的去乙酰化酶,参与基因沉默、细胞周期、炎症、氧化应激等过程。近来多项研究证实SIRT1参与抑郁症的发病,本文对SIRT1与抑郁症的关系进行综述,并从神经炎症、神经发生、昼夜节律、内质网应激等方面概括SIRT1参与抑郁症的可能机制,以期为抑郁症的治疗提供潜在的靶点。

ABSTRACT

Depression is a common mental illness, its etiology and mechanism are not fully elucidated, leading to poor therapeutic effect. Silencing information regulator 2 related enzyme 1(Sirtuin 1, SIRT1), an NAD+ dependent deacetylase, involved in the process of gene silencing, cell cycle, inflammation and oxidative stress. Recently numerous studies have confirmed that SIRT1 is involved in the pathogenesis of depression. In this review, we clarify the relationship between SIRT1 and depression, and its possible mechanism from aspects of neuroinflammation, neurogenesis, circadian rhythm and endoplasmic reticulum stress, so as to provide a potential target for the treatment of depression.
