【摘要】
目的 重性抑郁障碍（Major Depressive Disorder,MDD） 是严重的精神疾病，具有较高的自杀风险。本研究旨在筛选自杀未遂MDD患者的关键基因并评估其诊断价值。

方法 利用基因表达数据库（Gene Expression Omnibus,GEO）中的GSE247998数据集，分析27名健康对照、23名自杀未遂MDD患者和29名不伴自杀未遂MDD患者的外周血转录组数据。通过R语言筛选差异表达基因，使用STRING数据库构建蛋白质-蛋白质相互作用网络，并通过Cytoscape软件识别关键基因。利用pROC包计算关键基因的受试者工作特征（Receiver Operating Characteristic,ROC）曲线下面积（Area Under the Curve,AUC），评估其诊断价值。

结果 与健康对照相比，自杀未遂MDD患者中筛选出42个差异基因（11个上调，31个下调），识别出关键基因MMP8和LCN2，其AUC值分别为0.71和0.68。与不伴自杀未遂MDD患者相比，自杀未遂MDD患者中筛选出10个差异基因（1个上调，9个下调），识别出关键基因FANCA、CDC20和IFI27，其AUC值分别为0.78、0.76和0.58。

结论MMP8和LCN2在区分自杀未遂MDD患者和健康对照中具有诊断潜力，而FANCA和CDC20在区分自杀未遂和不伴自杀未遂的MDD患者中具有潜在诊断价值。

ABSTRACT

【【Abstract】
Objective Major depressive disorder (MDD) is a severe mental illness associated with a high risk of suicide. This study aims to identify hub genes in MDD patients with suicide attempts and evaluate their diagnostic value.

Methods Using the GSE247998 dataset from the Gene Expression Omnibus (GEO) database, we analyzed peripheral blood transcriptome data from 27 healthy controls, 23 MDD patients with suicide attempts, and 29 MDD patients without suicide attempts. Differentially expressed genes were screened using R, and a protein-protein interaction network was constructed. Hub genes were identified using Cytoscape software. The diagnostic value of these hub genes was evaluated by calculating the area under the receiver operating characteristic (ROC) curve (AUC) using the pROC package.

Results Compared to healthy controls, 42 differentially expressed genes (11 upregulated, 31 downregulated) were identified in MDD patients with suicide attempts. The hub genes MMP8 and LCN2 had AUC values of 0.71 and 0.68, respectively. Compared to MDD patients without suicide attempts, 10 differentially expressed genes (1 upregulated, 9 downregulated) were identified in MDD patients with suicide attempts. The hub genes FANCA, CDC20, and IFI27 had AUC values of 0.78, 0.76, and 0.58, respectively.

Conclusion   MMP8 and LCN2 have diagnostic potential for distinguishing MDD patients with suicide attempts from healthy controls, while FANCA and CDC20 have potential diagnostic value for distinguishing MDD patients with and without suicide attempts.
